Da bi to dosegli, je treba pridobiti \u010dim bolj\u0161i vpogled v biotsko raznovrstnost travnikov. V ta namen uporabljamo <b><a href=\"http:\/\/www.si-hr.eu\/2127\/2025\/03\/17\/biotska-raznovrstnost-suhih-travisc-2024-bioraznolikost-suhih-travnjaka-2024-prvi-rezultati-klasicna-metoda\/\">klasi\u010dne metode*<\/a><\/b>, v zadnjem \u010dasu pa vse pogosteje uporabljamo tehniko <b>metabarkodiranja DNA.<\/b><\/p>\n<\/p>\n<p><b>KAJ JE METABARKODIRANJE DNA?<\/b><\/p>\n<p align=\"justify\"><b>Metabarkodiranje DNA<\/b> je sodoben laboratorijsko-ra\u010dunalni\u0161ki pristop, ki iz vzorca analizira specifi\u010dna zaporedja DNA za posamezne vrste. Dolo\u010danje vrst poteka s primerjavo z zaporedji, shranjenimi v referen\u010dnih bazah podatkov. Na ta na\u010din se dolo\u010di sestava in raznolikost celotnega vzorca. Za razliko od klasi\u010dnega pristopa ta metoda omogo\u010da so\u010dasno dolo\u010danje vseh vrst nevreten\u010darjev, prisotnih v vzorcu. To omogo\u010da hiter in natan\u010den vpogled v biotsko raznovrstnost, brez potrebe po anga\u017eiranju taksonomskih specialistov za posamezne skupine. Da bismo to postigli, potrebno je najprije ste\u0107i \u0161to bolji uvid u bioraznolikost promatranog podru\u010dja. U tu svrhu koristimo <\/span><b style=\"font-size: 16.5px\" data-fusion-font=\"true\"><a href=\"http:\/\/www.si-hr.eu\/2127\/2025\/03\/17\/biotska-raznovrstnost-suhih-travisc-2024-bioraznolikost-suhih-travnjaka-2024-prvi-rezultati-klasicna-metoda\/\">klasi\u010dne metode*<\/a>,<\/b><span style=\"font-size: 16.5px\" data-fusion-font=\"true\"> ali u novije vrijeme sve vi\u0161e i tehniku <\/span><strong style=\"font-size: 16.5px\" data-fusion-font=\"true\">DNA metabarkodiranja<\/strong><span style=\"font-size: 16.5px\" data-fusion-font=\"true\">.<\/span><\/i><\/p>\n<\/p>\n<p><strong style=\"font-size: 16.5px\" data-fusion-font=\"true\">\u0160TO JE DNA METABARKODIRANJA?<\/strong><\/p>\n<p align=\"justify\"><i><strong style=\"font-size: 16.5px\" data-fusion-font=\"true\">DNA metabarkodiranje<\/strong><span style=\"font-size: 16.5px\" data-fusion-font=\"true\"><span> <\/span>je suvremeni laboratorijsko-ra\u010dunalni pristup kojim se iz prikupljenog uzorka analiziraju DNA sljedovi specifi\u010dni za pojedine vrste, a njihovo se odre\u0111ivanje provodi usporedbom sa sekvencama pohranjenima u referentnim bazama podataka \u010dime se utvr\u0111uje sastav i raznolikost \u010ditave zajednice. Za razliku  od klasi\u010dnog pristupa, ova metodologija omogu\u0107uje simultano odre\u0111ivanje vrsta beskralje\u0161njaka prisutnih u uzorku \u010dime se dobiva  brz i precizan uvid u bioraznolikost, bez potrebe za anga\u017emanom taksonomskih specijalista za pojedine skupine. Mogu\u0107nost i pouzdanost odre\u0111ivanja vrsta, me\u0111utim, uvelike ovise o postojanju i cjelovitosti <\/span><strong style=\"font-size: 16.5px\" data-fusion-font=\"true\">baza podataka<\/strong><span style=\"font-size: 16.5px\" data-fusion-font=\"true\"><span>,<\/span> pri \u010demu je za njihovu izgradnju i pouzdanost klju\u010dan doprinos fusion-text-3\"><p><b>METODA DNA METABARKODIRANJA V PROJEKTU CAREX<\/b><\/p>\n<p align=\"justify\">Cilj projekta CarEx je pridobiti <b>\u0161irok vpogled v biotsko raznovrstnost<\/b> nevreten\u010darjev suhih travnikov na obmo\u010dju Kozjanskega parka in parka prirode \u017dumberak Samoborsko gorje. Ocena biotske raznovrstnosti se obi\u010dajno izvaja z analizo izbranih skupin rastlin ali \u017eivali, za katere so na voljo strokovnjaki s potrebnim znanjem o dolo\u010danju vrst. Ta pristop se imenuje <b><span>klasi\u010dna metoda<\/span>.<\/b> Zahteva delo obi\u010dajno \u010dasovno zelo obremenjenih strokovnjakov  &#8211; taksonomov,, postopek pridobivanja rezultatov je dolgotrajen in kompleksen.<\/p>\n<p align=\"justify\">Z uporabo <b><span>DNA metabarkodiranja<\/span> <\/b>v projektu bomo biotsko raznovrstnost opisali veliko hitreje in bolj celovito, hkrati bomo raziskali tudi sezonske spremembe v sestavi zdru\u017eb, kot tudi vplive razli\u010dnih na\u010dinov upravljanja. Zbrani podatki nam bodo omogo\u010dili razvoj novih re\u017eimov upravljanja, ki jih testiramo na poskusnih parcelah projekta. Procjena stanja bioraznolikosti uobi\u010dajeno se provodi analizom odabranih skupina biljaka ili \u017eivotinja za koje su dostupni stru\u010dnjaci s potrebnim znanjem identifikacije vrsta. Taj se pristup naziva <\/span><strong style=\"font-size: 16.5px\" data-fusion-font=\"true\">klasi\u010dnom metodom <\/strong><span style=\"font-size: 16.5px\" data-fusion-font=\"true\">koja zahtijeva te\u0161ko dostupne specijalizirane taksonome, dok je sam proces dobivanja rezultata dugotrajan i slo\u017een.<\/span><\/i><\/p>\n<p align=\"justify\"><i><span style=\"font-size: 16.5px\" data-fusion-font=\"true\">Primjenom <\/span><strong style=\"font-size: 16.5px\" data-fusion-font=\"true\">DNA metabarkodiranja<\/strong><span style=\"font-size: 16.5px\" data-fusion-font=\"true\"><span> <\/span>u projektu stoga mo\u017eemo znatno br\u017ee i obuhvatnije opisati bioraznolikost, uklju\u010duju\u0107i sezonske promjene, kao i izravne u\u010dinke razli\u010ditih na\u010dina upravljanja na zajednice organizama. Novoprikupljeni podaci omogu\u0107it \u0107e razvoj i testiranje pionirskih re\u017eima upravljanja (s naglaskom na ko\u0161nju) na pokusnim plohama projekta.
